This is a working overview of Semax, written for readers who want more than a one-paragraph summary but less than a textbook.
This page was last updated on 2026-05-13 and is reviewed periodically as new material appears.
Semax is a synthetic heptapeptide with the sequence Met-Glu-His-Phe-Pro-Gly-Pro. Its chain combines the first seven residues of corticotropin with a C-terminal proline-glycine-proline extension, a modification intended to slow enzymatic breakdown. The free peptide has a molecular mass near 813.9 daltons. It belongs to the class of ACTH-derived fragments studied for central nervous system activity rather than for adrenal steroid stimulation. This structural relationship to a natural hormone fragment is the usual starting point for describing the compound in the literature.
Development took place during the 1980s at institutes of the Russian Academy of Sciences, where peptide fragments of ACTH were screened for cognitive and neuroprotective effects. The compound received regulatory approval in Russia as a nasal preparation, marketed for conditions such as ischemic stroke, transient ischemic attacks, and optic nerve disorders. Registration in other countries has not followed. Most published clinical reports originate from a small number of Russian research groups, and independent replication outside that setting remains limited. The regulatory status therefore differs sharply between Russia and the rest of the world.
Routine characterization relies on reversed-phase high-performance liquid chromatography to establish purity and on mass spectrometry to confirm molecular identity. Electrospray ionization and matrix-assisted laser desorption ionization are both used for mass verification. Amino acid analysis and peptide mapping can detect sequence errors. Common impurities include truncated sequences, methionine sulfoxide formed by oxidation, and deamidated products. Chromatograms are usually recorded near 214 nm, where the peptide backbone absorbs, and purity is reported as the percentage area of the principal peak.
Verification of a supplied batch generally combines a certificate of analysis with independent testing, because certificates are self-reported documents. A typical package includes a chromatographic trace, a mass spectrum, and a stated water or counter-ion content. Batch-to-batch consistency matters more than a single purity figure when results are compared across experiments. No single mandatory standard governs research-grade peptide release, so laboratories are expected to define their own acceptance criteria. Residual trifluoroacetate from purification is a frequently overlooked counter-ion.
Lyophilized material is chemically stable for extended periods when kept dry, cold, and protected from light. The powder is hygroscopic, so vials should be warmed to room temperature before opening to reduce condensation on the contents. Once dissolved, the peptide is far less stable because peptide bonds are susceptible to hydrolysis and the methionine residue can oxidize. Solutions are typically aliquoted and held at 2-8 °C for short intervals or frozen for longer ones, and repeated freeze-thaw cycles should be avoided.

Published studies are dominated by animal models of stroke, ischemia, and cognitive impairment, with a smaller number of human trials conducted in Russia. Many of the human reports are small, single-center, and published in Russian-language journals, which limits independent scrutiny. Outcome measures vary between studies and often rely on clinician-rated scales rather than objective biomarkers. Systematic reviews have noted the methodological weaknesses and called for larger, preregistered trials before firm conclusions can be drawn.
Claims about enhanced focus, memory, or mood in healthy people rest mostly on anecdotal reports and community discussion rather than on controlled data. It remains unclear whether any cognitive benefit observed in patients recovering from brain injury would extend to uninjured users. Dose-response relationships, long-term safety, and interactions with other drugs are not well characterized in the peer-reviewed literature. Questions about optimal route of administration and treatment duration likewise remain open.
Scientific literature on semax is unevenly distributed. A substantial share of published work originates from a small number of laboratories in Russia, while independent replication elsewhere is limited. Human data consist mostly of small trials with short follow-up, and several reported outcomes rely on subjective rating scales. Questions about how much intact peptide reaches the central nervous system after nasal administration, and how long it persists there, are still unresolved. The compound is best described as an active research subject rather than a settled pharmacological agent.
Semax is a synthetic heptapeptide with the sequence Met-Glu-His-Phe-Pro-Gly-Pro. It was derived from the ACTH(4-10) fragment, a short segment of adrenocorticotropic hormone that lacks the hormonal activity associated with the full-length peptide. Researchers at the Institute of Molecular Genetics in Moscow developed the compound during the 1980s. It has been registered as a pharmaceutical product in Russia and several neighbouring countries, where it is supplied as a nasal solution, and it is also sold internationally as a research chemical.
=== Natural occurrence === Because of the short half-life of all known isotopes of fermium, any primordial fermium, that is fermium present on Earth during its formation, has decayed by now. Synthesis of fermium from naturally occurring uranium and thorium in the Earth's crust requires multiple neutron captures, which is extremely unlikely. Therefore, most fermium is produced on Earth in laboratories, high-power nuclear reactors, or in nuclear tests, and is present for only a few months afterward. The transuranic elements up to fermium should have been present in the natural nuclear fission reactor at Oklo, but any quantities produced then would have long since decayed away.
==== Switzerland ==== While THC remains illegal, cannabidiol is not subject to the Swiss Narcotic Acts because it does not produce a comparable psychoactive effect. Cannabis products containing less than 1% THC can be sold and purchased legally.
== Early life and education == Vasquez was born on August 3, 1984, in El Paso, Texas. He was raised in the El Paso–Juárez–Las Cruces border region, spending much of his childhood in both the United States and Mexico. He grew up in Ciudad Juárez, Mexico, where he spent time in his grandfather's television repair shop. His grandfather Javier Bañuelos, originally from Zacatecas, Mexico, had transitioned from farming to working as a mailman before establishing his repair business. Vasquez later attended Montwood High School in El Paso and then enrolled at New Mexico State University (NMSU) in Las Cruces, where he earned a Bachelor of Arts degree in English and journalism in 2008. During college, he served as the news editor and later as editor-in-chief of The Round Up, NMSU's student-run newspaper.
Conversely, a decrease in temperature results in a decrease in respiratory pigment cooperativity and increase in affinity. The slight rise in P50 that occurs with temperature change allows oxygen pressure to remain high in the capillaries, allowing for elevated diffusion of oxygen into the mitochondria during periods of high oxygen consumption. The increase in temperature results in higher enzyme activity, yet the decrease in hemocyanin affinity allows enzyme activity to remain constant and maintain homeostasis. The highest hemolymph protein concentrations are seen at 32 °C (90 °F) and then drop at temperatures above this. Oxygen affinity in the blood decreases by 0.20 kPa/°C (0.016 psi/°F) at a pH of 7.4. The octopod's thermal tolerance is limited by its ability to consume oxygen, and when it fails to provide enough oxygen to circulate at extreme temperatures the effects can be fatal. O. vulgaris has a pH-independent venous reserve that represents the amount of oxygen that remains bound to the respiratory pigment at constant pressure of oxygen. This reserve allows the octopus to tolerate a wide range of pH related to temperature. As a temperature conformer, O. vulgaris does not have any specific organ or structure dedicated to heat production or heat exchange. Like all animals, they produce heat as a result of ordinary metabolic processes such as digestion of food, but take no special means to keep their body temperature within a certain range. Their preferred temperature directly reflects the temperature to which they are acclimated.

Fatty foods are sensitive to photooxidation, which forms hydroperoxides by oxidizing unsaturated fatty acids and ester. Exposure to ultraviolet (UV) radiation can cause direct photooxidation and decompose peroxides and carbonyl molecules. These molecules undergo free radical chain reactions, but antioxidants inhibit them by preventing the oxidation processes.
=== Screening === Two main approaches exist for the finding of new bioactive chemical entities from natural sources. The first is sometimes referred to as random collection and screening of material, but the collection is far from random. Biological (often botanical) knowledge is often used to identify families that show promise. This approach is effective because only a small part of the earth's biodiversity has ever been tested for pharmaceutical activity. Also, organisms living in a species-rich environment need to evolve defensive and competitive mechanisms to survive. Those mechanisms might be exploited in the development of beneficial drugs. A collection of plant, animal and microbial samples from rich ecosystems can potentially give rise to novel biological activities worth exploiting in the drug development process. One example of successful use of this strategy is the screening for antitumor agents by the National Cancer Institute, which started in the 1960s. Paclitaxel was identified from Pacific yew tree Taxus brevifolia. Paclitaxel showed anti-tumour activity by a previously undescribed mechanism (stabilization of microtubules) and is now approved for clinical use for the treatment of lung, breast, and ovarian cancer, as well as for Kaposi's sarcoma. Early in the 21st century, Cabazitaxel (made by Sanofi, a French firm), another relative of taxol has been shown effective against prostate cancer, also because it works by preventing the formation of microtubules, which pull the chromosomes apart in dividing cells (such as cancer cells). Other examples are: 1.
==== Two-compartment model ==== Not all body tissues have the same blood supply, so the distribution of the drug will be slower in those tissues than in others with a better blood supply. Furthermore, there are some tissues (such as the brain tissue) that present a real barrier to the distribution of drugs, which may be breached with greater or lesser ease depending on the drug's characteristics. If these relative conditions for the different tissue types are considered along with the rate of elimination, the organism can be considered to be acting like two compartments: one that we can call the central compartment, which has a more rapid distribution and consists of organs and systems with a well-developed blood supply; and the peripheral compartment, which is made up of organs with a lower blood flow. Other tissues, such as the brain, can occupy a variable position depending on a drug's ability to passively transport (high lipophilicity) and evade active efflux to cross the blood–brain barrier (BBB) that separates the organ from the blood supply. Two-compartment models vary depending on which compartment elimination occurs in. The most common situation is that elimination occurs in the central compartment as the liver and kidneys are organs with a good blood supply. However, in some situations, elimination occurs in the peripheral compartment or even in both compartments. This can mean that there are three possible variations in the two compartment model, which still do not cover all possibilities.
== History == The company was founded in 1929 by Dharampal Sugandhi in Chandni Chowk, Delhi, as a perfumery business producing incense sticks, rose water, and tobacco products. In 1958, Satyapal Sugandhi introduced the BABA tobacco brand. During the 1980s, the group diversified into packaged foods with the launch of Catch Salt & Pepper (1987), followed by Catch Natural Spring Water in 1999. The group's subsequent launches included confectionery products under brands such as Rajnigandha, Pass Pass, and Pulse Candy. In 2019, the DS Group acquired an 11–18% stake in the Switzerland-based Lalique Group . In 2023, it introduced the Swiss chocolate brand Läderach to India and acquired the Indian confectionery brand LuvIt.
Following independence on 15 August 1947, Jawaharlal Nehru served as prime minister of the Dominion of India, remaining caretaker until 1951, shortly after India became a republic in 1950. Led by Nehru, the Congress won the 1951, 1957, and 1962 general elections comfortably. After Nehru died in May 1964, Lal Bahadur Shastri became prime minister. Shastri died in January 1966, shortly after signing the Tashkent Peace Declaration. The Congress chose Indira Gandhi as prime minister, leading the party to election victories in 1967 and 1971. In 1975, she declared a state of emergency, suspending civil liberties, which led to Congress being voted out in 1977. Congress returned to power in 1980. Following a military operation at the Golden Temple in Amritsar, Indira Gandhi was assassinated on 31 October 1984. Indira Gandhi's son, Rajiv Gandhi, succeeded her and won the late 1984 elections. During the general election campaign of 1991, Rajiv Gandhi was assassinated by a Sri Lankan Tamil separatist. Congress emerged from the elections as the largest party, and P. V. Narasimha Rao formed a minority government that completed a full five-year term. After the 1999 elections, the BJP's Atal Bihari Vajpayee led a National Democratic Alliance (NDA) coalition and became the first non-Congress prime minister to complete a five-year term. The NDA was defeated in 2004 by the Congress-led United Progressive Alliance (UPA). Manmohan Singh served as prime minister with external support, returning with increased, independent numbers in 2009.

== Overview == The RM-ODP is a reference model based on precise concepts derived from current distributed processing developments and, as far as possible, on the use of formal description techniques for specification of the architecture. Many RM-ODP concepts, possibly under different names, have been around for a long time and have been rigorously described and explained in exact philosophy (for example, in the works of Mario Bunge) and in systems thinking (for example, in the works of Friedrich Hayek). Some of these concepts—such as abstraction, composition, and emergence—have recently been provided with a solid mathematical foundation in category theory. RM-ODP has four fundamental elements:
==== Travelling wave ion mobility spectrometry (TWIMS) ==== In TWIMS, ions are separated according to their mobility through a travelling wave in a gas filled cell. Both radio-frequency (RF) and direct current (DC) voltages are applied to a series of ring electrodes called a stacked ring ion guide (SRIG) to confine the ions and create a travelling wave. Based on the speed and magnitude of the travelling wave, ions can be separated. Smaller ions have higher mobility through the wave due to fewer collisions with gas molecules and exit the cell faster than ions of lower mobility (larger ions). Similar to DTIMS, CCS values of ions can be calculated with TWIMS using a calibration derived with known standards. A commercial example of the TWIMS-MS instrumentation is Waters Corp Synapt G2-S instrument.
=== Pharmacokinetics === Topical hydrocortisone has minimal absorption into the body: Only 4-19% of the topical hydrocortisone cream applied would be absorbed into the human bloodstream. After a certain extent of absorption, it undergoes distribution, metabolism and elimination pathways that are similar to systemic hydrocortisone. Regarding the distribution of topical hydrocortisone, it binds to plasma proteins such as globulin and albumin, then the drug is mainly metabolized in the liver, and the metabolite will be excreted through bile or by kidneys.
Suppress the release of neuropeptide Y (NPY), which in turn prevents the release of appetite enhancing orexins from the lateral hypothalamus. This decreases appetite and food intake, promoting weight loss. Stimulate the expression of cocaine and amphetamine regulated transcript (CART). Though rising blood levels of leptin do promote weight loss to some extent, its main role is to protect the body against weight loss in times of nutritional deprivation. Other factors also have been shown to effect long-term hunger and food intake regulation including insulin. In addition, the biological clock (which is regulated by the hypothalamus) stimulates hunger. Processes from other cerebral loci, such as from the limbic system and the cerebral cortex, project on the hypothalamus and modify appetite. This explains why in clinical depression and stress, energy intake can change quite drastically.

Semax is a synthetic seven-amino-acid peptide derived from a fragment of corticotropin. It is used in Russia as a nasal preparation, while elsewhere it is studied as a research compound. It is not approved as a medicine in most countries.
Its sequence corresponds to the 4-10 region of corticotropin extended by three residues. This fragment lacks the region responsible for adrenal steroid release, so it is explored for neural rather than hormonal effects. The relationship explains the compound's classification among ACTH-derived peptides.
It holds approval in Russia for several neurological indications as a nasal spray or drop formulation. Regulatory agencies in the United States and the European Union have not approved it. Products sold elsewhere are generally labeled for research use only.
Dry powder is normally held at -20 °C or lower, away from light and moisture. Sealed vials can also be kept at 2-8 °C for shorter intervals. Warming to room temperature before opening prevents condensation.
