RP-HPLC raises a handful of sensible questions. This page answers them in order, starting with the fundamentals and moving to applications.
Reviewed 2026-06-13. Anything still debated is marked as such rather than presented as settled.
Published research has focused mainly on neurological and cognitive endpoints in animal models, with proposed mechanisms involving brain-derived neurotrophic factor and related signalling pathways. A substantial share of the human data originates from a limited number of research groups, and independent replication in other countries remains sparse. Regulatory status reflects that distribution: the peptide is registered as a medicine in Russia and appears in some neighbouring markets, while elsewhere it is handled as a research chemical without approved therapeutic labelling. Questions about dose-response relationships, long-term effects, and comparability across studies are still open.
Lyophilised powder is normally kept at -20 °C in a desiccated container, with some suppliers recommending -80 °C for long-term archival storage. Repeated freeze-thaw cycles are the most common cause of avoidable loss, so aliquoting before freezing reduces variability between working sessions. Dissolved peptide is far less stable than the dry solid and is usually prepared fresh or held briefly at 4 °C. Aqueous solutions support both hydrolysis of the backbone and oxidation of the N-terminal methionine, and these two routes dominate degradation under ordinary laboratory conditions.
Identity and purity are confirmed with reversed-phase high-performance liquid chromatography, typically monitored at 214 nanometres where the peptide bond absorbs. Mass spectrometry, either electrospray or MALDI-TOF, verifies molecular mass against the theoretical value and detects truncation or adduct formation. Amino acid analysis and peptide mapping provide additional confirmation when required. The most frequently reported impurities are deletion sequences from incomplete coupling, methionine sulfoxide from oxidation, and dimeric species formed through non-covalent aggregation. Impurity profiles depend strongly on the synthesis and purification route chosen by the producer.
Later generations of the molecule include an N-acetylated form and an amidated form, both marketed online alongside the parent peptide. These variants differ in terminal chemistry and stability, and they are frequently discussed in the same breath even though they have not been compared in controlled trials. Supply outside formal healthcare systems comes largely from laboratories that synthesize peptides to order. Purity and identity of these materials vary widely, and no single body oversees the international trade.
Semax is a synthetic heptapeptide developed in the Soviet Union during the 1980s by researchers at the Institute of Molecular Genetics in Moscow. It was designed as a truncated analog of adrenocorticotropic hormone, retaining only the fragment spanning residues four through ten. Investigators sought a peptide that would preserve the cognitive effects associated with ACTH while eliminating the hormonal stimulation of the adrenal cortex. The compound entered clinical use in Russia during the following decade.

Scientific literature on semax is unevenly distributed. A substantial share of published work originates from a small number of laboratories in Russia, while independent replication elsewhere is limited. Human data consist mostly of small trials with short follow-up, and several reported outcomes rely on subjective rating scales. Questions about how much intact peptide reaches the central nervous system after nasal administration, and how long it persists there, are still unresolved. The compound is best described as an active research subject rather than a settled pharmacological agent.
Semax is a synthetic heptapeptide with the sequence Met-Glu-His-Phe-Pro-Gly-Pro. It was derived from the ACTH(4-10) fragment, a short segment of adrenocorticotropic hormone that lacks the hormonal activity associated with the full-length peptide. Researchers at the Institute of Molecular Genetics in Moscow developed the compound during the 1980s. It has been registered as a pharmaceutical product in Russia and several neighbouring countries, where it is supplied as a nasal solution, and it is also sold internationally as a research chemical.
The proposed mechanism centres on neurotrophic signalling rather than direct receptor activation. Semax is reported to increase expression of brain-derived neurotrophic factor and nerve growth factor in several brain regions, and to shift the balance between excitatory and inhibitory neurotransmitter systems. Interaction with melanocortin receptors has been suggested because of the parent ACTH fragment. Many of these findings come from rodent studies, and the extent to which they translate to human physiology remains an open question.

== Bioinformatics tools == Bioinformatics tools exist to assist with interpretation of mass spectra (see de novo peptide sequencing), to compare or analyze protein sequences (see sequence analysis), or search databases using peptide or protein sequences (see BLAST).
== Epidemiology == A study from the Netherlands has shown an incidence of 1 in 20 million births. According to the Progeria Research Foundation, as of September 2020, there are 179 known cases in the world, in 53 countries; 18 of the cases were identified in the United States. Hundreds of cases have been reported in medical history since 1886. However, the Progeria Research Foundation believes there may be as many as 150 undiagnosed cases worldwide. There have been only two cases in which a healthy person was known to carry the LMNA mutation that causes progeria. One family from India had four of six children with progeria.
== Liquid chromatography == Back pressure is the term used for the hydraulic pressure required to create a flow through a chromatography column in high-performance liquid chromatography, the term deriving from the fact that it is generated by the resistance of the column, and exerts its influence backwards on the pump that must supply the flow. Back-pressure is a useful diagnostic feature of problems with the chromatography column. Rapid chromatography is favoured by columns packed with very small particles, which create high back-pressures. Column designers use "kinetic plots" to show the performance of a column at a constant back-pressure, usually selected as the maximum that a system's pump can reliably produce.
A gel refers to the semi-solid, 3-dimensional matrix formed from an interspersed system of colloidal particles or the permeation of a solvent into an entwined polymer chain network. Pharmaceutical gels are formed by adding a gelator (gelling agent) to the solvent and active ingredient mixture. Gelators used in gel formulation can be small molecules with low molecular weight or polymers (synthetic, semi-synthetic or natural). The solvent that is used as a dispersion medium can be aqueous, organic, inorganic, or a system of different solvents. Topical gels are used as a contact or transport medium for active drugs to act on or through the skin. The active drug molecules are entwined into the 3D mesh of the gel and delivered to the site of action.

==== United States ==== In July 2025, the US Food and Drug Administration (FDA) formally recommended that the Drug Enforcement Administration (DEA) classify 7-OH-MIT as a controlled substance. This action was publicized to not be targeting Mitragyna speciosa leaf powder itself. Despite claims by marketers for products that contain 7-OH-MIT that they can be used to treat anxiety and pain, the drug is not approved by the FDA for any medical use or as a food supplement.
Joseph Robinette Biden Jr. was born on November 20, 1942, at St. Mary's Hospital in Scranton, Pennsylvania, to Catherine Eugenia "Jean" Biden (née Finnegan) and Joseph Robinette Biden Sr. He is the oldest child in a Catholic family of predominantly Irish descent. Biden has a sister, Valerie, and two brothers, James and Francis. Joseph Sr. had been wealthy, and the family purchased a home in the affluent Long Island suburb of Garden City, New York, in 1946. After he suffered business setbacks around the time Biden was seven years old, the family lived with Jean's parents in Scranton for several years. Scranton fell into economic decline during the 1950s, and Joseph Sr. could not find steady work. Beginning in 1953, when Biden was ten, the family lived in an apartment in Claymont, Delaware, before moving to a house in nearby Mayfield, Delaware. Joseph Sr. later became a successful used-car salesman, maintaining the family in a middle-class lifestyle. Growing up, Biden observed alcoholism in his family, deciding at a young age to be teetotal. At Archmere Academy in Claymont, Biden played baseball and was a standout halfback and wide receiver on the high school football team. Though a poor student, he was class president in his junior and senior years. He graduated in 1961. At the University of Delaware in Newark, Biden briefly played freshman football and received a Bachelor of Arts degree with a double major in history and political science in 1965. To overcome a childhood stutter, he memorized lines from Ralph Waldo Emerson and William Butler Yeats.
A section of the report pertaining to the internal functioning of the S&D Group was held over until September 2023 to allow for a discussion of its findings with the S&D Bureau prior to its publication.

Reversed-phase HPLC gives the main purity figure, most often with UV detection near 214 nanometres. Mass spectrometry then confirms the molecular mass. Together the two methods distinguish a correct sequence from a closely related impurity.
The main chemical risks are methionine oxidation and backbone hydrolysis in solution. Moisture and repeated temperature cycling accelerate both processes. Dry powder held cold and desiccated is considerably more stable than any reconstituted preparation.
It holds a medicine registration in Russia, where it has been used clinically for decades. In most other countries it is treated as a research chemical. That split explains the uneven distribution of clinical literature.
It originated at the Institute of Molecular Genetics in Moscow during the 1980s. The work was carried out by a Russian research group that specialized in peptide neuropharmacology.
